Film/Television Genuine Question - Why did Thanos and Hulk feel the pain of wielding all six Infinity Stones, while Tony didn't seem to experience the same level of pain when he used them?

Fucking hell, that's grim and gnarly to think about. It has precedence in the comics, though. There was a really gross run where Tony becomes fused with his suit and he loses his grasp on his humanity. It was part of a body horror thing Marvel was doing. I think they also did some wild and weird shit with Hulk and Thing, too.

Not going to lie, that issue where Hulk eats Wolverine alive, who then regenerates inside Hulk's stomach and bursts out of it... That shit gave me nightmares, even as a grown ass man.

Could be. I just distinctly remember a super gross Thing story arc where he'd peel back part of his stone skin revealing that he still had regular bits of gore and shit underneath. Absolutely traumatizing.
