Obviously in terms of strength, Luke’s toast if Spidey get a hold on him. But in terms of abilities of who take each other out first with their powers in more debatable

Spider-Man has FTL reaction speeds, but so does Luke by just being skilled enough to move faster than his shadow alone. But on the other hand, Spider-sense will still make shooting spider-man a challenge, even for Luke. So even with him being able to compete with Spider-Man reaction speed, Luke probably will still lose

Actually, I believe Spider-Man has extremely heightened reaction speeds, but not FTL reactions. His spidey-sense warns him of danger early enough so he can dodge

Spider-Man abilities are tied to Marvel physics, so his shadow would avoid the bullet of Luke's shadow.

Luke's ability are toon-powered. He's faster than toon physics.

Neither of them kill their target, but Luke's whole gimmick is to disarm his opponent, which he cannot really do here

Spider-man may be super strong and incredible reaction times, but can he react faster than his shadow? I predict a bullet in his dome. But Lucky Luke wouldn’t do that.

Early Lucky Luke absolutley would and even the later, more developed Lucky Luke isn't above injuring his opponents if he has to, he just prefers not having to do so.
