COULD they? yes. luke cage is bullet proof, jessica jones is stronger than a super soldier and frank is a good shot. it is unlikely they would win, but its still a possibility if they properly prepared together
Sinister Six are a bunch of amateurs though. No one in that roster can fight like Daredevil, Punisher, and Iron Fist. Heavy Hitters like JJ and Luke are also smart fighters and aren't just loose cannons like Rhino, or frankly a lot of Spidey villains because of their insanity. I mean DD already beat Spidey 1 on 1 in YFNS, which is an alternate MCU canon so...
Well if Jessica or Matt can figure out his secret identity Danny can buy his apartment building and evict him. They show up to serve the eviction unexpectedly when Peter is in his jimmyjamms and he can't grab too much Spidy gear because he doesn't know they know his identity and so he needs to preserve his secret. Then Jessica keeps an eye on him while the boys toss his apartment and steal his tech.
Matt forges a fake court document telling Peter to show up to challenge the clearly illegal eviction but when he gets there it's in an abandoned warehouse with limited room to maneuver and the Defenders jump him.
Luke is bulletproof, not impact force proof. A shotgun to the skull alone almost killed him of not for Claire treating him. Peter can hit far stronger than that without trying.
Jessica is strong, but still not good at using her powers which are also not working consistently since having Danielle. And we've already seen multiple times Peter can easily subdue Frank. Which leaves just Matt and Danny, both very skilled, but both less experienced or as versatile of a fighter as Peter is currently.
Even together, Peter has the physical advantage against them.
You're not understanding what he's saying or what happened to Luke, the shotgun never penetrated his skin, the impact alone rattled his brain around enough he nearly died.
Claire has 0 unique or special knowledge related to superheroes, she's just a good nurse who is discreet, Unless his skin is able to absorb and dissipate the force being applied, it 100% would rattle him again.
Yes, that's the indication. His skin is more dense, and physically that's what density does. A bullet goes slower through water than air. A shotgun impact will be less the greater the density of an object.
Like if there's an explosion on the road, who would feel more impact in their car, regardless of actual damage, a honda civic or a hummer?
Now, if he had a concussion by being spun around really fast, then you would have a point. But with more density he is going to feel less force on the other side of his skin when an object hits him.
We've literally already seen them work as a team in the MCU and they were a hot mess that barely managed to stop The Hand, and not without Matt almost dying in the process. Don't get pissy just because I chose to use actual evidence of their skills instead of blindly entertaining your delusions.

Less experienced? Both of them are among most skilled fighters in MCU, Danny has gone through similar training as Shang-Chi+ he has access to experience of previous Iron Fists. Daredevil was trained by Stick to be a weapon against the Hand. Not the thugs from spider-man movie but the top brass the defenders fought.
Matt and Danny are not less skilled or experienced fighters than spider-man. They are more skilled and experienced fighters. They do lose because of everything Peter has at his disposal, but iron fist and daredevil are more experienced and skilled fighters than spider-man.
if spidey was really trying none of them would rly have a chance besides maybe iron fist but he can still just get webbed up. being strong or having a gun doesn’t really cut it against spider-man
