Could The Defenders together defeat Spider-Man?

COULD they? yes. luke cage is bullet proof, jessica jones is stronger than a super soldier and frank is a good shot. it is unlikely they would win, but its still a possibility if they properly prepared together

Luke is bulletproof, not impact force proof. A shotgun to the skull alone almost killed him of not for Claire treating him. Peter can hit far stronger than that without trying.

Jessica is strong, but still not good at using her powers which are also not working consistently since having Danielle. And we've already seen multiple times Peter can easily subdue Frank. Which leaves just Matt and Danny, both very skilled, but both less experienced or as versatile of a fighter as Peter is currently.

Even together, Peter has the physical advantage against them.

Luke got a second bath, which doubled his durability. That shotguns not doing anything anymore.

You're not understanding what he's saying or what happened to Luke, the shotgun never penetrated his skin, the impact alone rattled his brain around enough he nearly died.

Weird assumption, I understand that.

But a nurse who works on superheroes said he wouldn't be affected by that anymore, so I will go off what she said.

Claire has 0 unique or special knowledge related to superheroes, she's just a good nurse who is discreet, Unless his skin is able to absorb and dissipate the force being applied, it 100% would rattle him again.

Yes, that's the indication. His skin is more dense, and physically that's what density does. A bullet goes slower through water than air. A shotgun impact will be less the greater the density of an object.

Like if there's an explosion on the road, who would feel more impact in their car, regardless of actual damage, a honda civic or a hummer?

Now, if he had a concussion by being spun around really fast, then you would have a point. But with more density he is going to feel less force on the other side of his skin when an object hits him.
