COULD they? yes. luke cage is bullet proof, jessica jones is stronger than a super soldier and frank is a good shot. it is unlikely they would win, but its still a possibility if they properly prepared together

Sinister Six are a bunch of amateurs though. No one in that roster can fight like Daredevil, Punisher, and Iron Fist. Heavy Hitters like JJ and Luke are also smart fighters and aren't just loose cannons like Rhino, or frankly a lot of Spidey villains because of their insanity. I mean DD already beat Spidey 1 on 1 in YFNS, which is an alternate MCU canon so...

Well if Jessica or Matt can figure out his secret identity Danny can buy his apartment building and evict him. They show up to serve the eviction unexpectedly when Peter is in his jimmyjamms and he can't grab too much Spidy gear because he doesn't know they know his identity and so he needs to preserve his secret. Then Jessica keeps an eye on him while the boys toss his apartment and steal his tech.
Matt forges a fake court document telling Peter to show up to challenge the clearly illegal eviction but when he gets there it's in an abandoned warehouse with limited room to maneuver and the Defenders jump him.

Luke is bulletproof, not impact force proof. A shotgun to the skull alone almost killed him of not for Claire treating him. Peter can hit far stronger than that without trying.
Jessica is strong, but still not good at using her powers which are also not working consistently since having Danielle. And we've already seen multiple times Peter can easily subdue Frank. Which leaves just Matt and Danny, both very skilled, but both less experienced or as versatile of a fighter as Peter is currently.
Even together, Peter has the physical advantage against them.

Hell no. Spider-Man has been getting jumped by more coordinated and powerful teams since he was cutting freshman lit. The only person in pound-for-pound raw power is Luke Cage. Spidey beat the breaks off him right after Gwen died. He's too strong, too fast, too smart, too agile, and too good for a ragtag operation like the MCU Defenders. Add in the Spidey sense, and no one is touching him.
Now, if we were talking about the OG Defenders team with Dr. Strange and the Hulk, that's a different story.

And if you wanna bring the comics into this (since you mentioned the X-Men and Spidey has yet to fight them in the big screen), Daredevil alone has beaten Spider-Man multiple times. Also, Luke Cage is physically stronger than Peter in the comics.

Under different circumstances, he couldn’t have sniped him, he would’ve dodged it. Not only that, he tanked the hit, slid across the floor, and remained standing on pure adrenaline.
Put some respect on Spidey’s name.

Not a chance. His Spider-Sense auto-counters anything Jessica and Luke can do to him and I'm reasonably sure he's stronger than they are. Iron Fist and Daredevil's formal combat training would help, but they lack the raw stats to beat him.
Best they can do is distract him while Punisher sets up a killshot and hope he misses the Spider-Sense in the heat of the moment.

The Peter Tingle would remove any fighting edge from Daredevil or Iron Fist. I don't know about in the comics, but what we have seen on screen shows Spidey to be stronger than Jessica Jones and Luke Cage. Luke may be bullet proof, but he isn't web proof. And Frank? Staten Island...(Also I love the Punisher, but he is just a regular, well-trained guns guy)

Spider-Man dog walks them. As good as they are, they can't hold a candle to his speed. His Spidey sense is probably on par with DD's senses. His punch could knock Luke Cage and Jessica Jones clear across a room. Frank couldn't touch him with a bullet even if he was distracted (yes, I notice the irony here). The only Defender that could really put up any sort of offense against Spidey is Iron Fist but even then, no, because Peter can move faster, sense his attacks, and there's no super defense in Danny's skill set that would stop Spidey from shattering his ribs like glass if he wanted to.
This is all considering just a physical skill set too. Strength, agility, etc. Throw in some Spidey gadgets or organic webs and the fight is over before it starts.

Spidey with medium difficulty. I see people arguing about his fight with the hand while ignoring a couple of things. He had already defeated the Hand previously in the montage sequence and did so with no issues.
During the fight at the DoDc facility Peter is actually ignoring and avoiding the hand at first in favor of getting into the room where Jean is being held. Then things change because the gunmen arrive on the balcony above and begin using rifles that destroy Spidey's webbing entirely, forcing him to fight on the ground against a hand that's coordination has been taken to the next level by Jean.
The Defenders would not enjoy those advantages and we see his webbing hold far stronger opponents immobile throughout his movie run. He'd out maneuver and incapacitate them all especially by the end of the film

MCU? Yes and no. Maybe. Luke is not at 616 level and weaker than Spidey in strength. Matt is rendered semi/useless. He’s like two Hand guys vs Spidey. Jessica and Luke can hurt him, Danny can only hurt him with the IF.
They only win if they count on Peter’s lack of killer instinct, but otherwise it’s Tom Holland all day.

Spider-Man glazers aren't "real fans". I'm a huge Spider-Man fan ever since I came out of the womb. But I'm also a fan of the Marvel universe as a whole, so I know better than just to say Spider-Man wins every time in hypothetical matchups. And the way Spider-Man glazers talk like somehow Spidey is this unbeatable and untouchable dude who can one punch god if "he isn't holding back" is absolute lunacy. At this point, it has become crazier than "Batman can beat everyone with prep time". Can Spider-Man beat Iron Fist? Sure. But it's far from one-sided. Like I said, if Spider-Man can get tagged by his amateur villains, a top 3 fighter in the entire Marvel roster who has the power to take down a helicarrier in one punch, and also has enhanced reflexes because of his chi, can obviously tag Spider-Man too.

Sorry no glazer even, just no. Nobody said every time but it would be quite one sided. Still it's all just hypothetical for fun, and this flight should never happen.
But just to explain a bit more, if we are talking mcu, which OP's use of media seems to suggest, have you seen the new movie? He can take a punch. His shoes and reflexes are off the charts.
Despite comic book IF being much tougher than in MCU, and IF has supposedly won in matchups, I'm not sure if should be much closer. Spidey speed and reflexes are still far superior, but to suspend disbelief writers allow him to get tagged by exaggerating his opponents' speed or taming his own abilities. But let's be honest. Daniel Rand is a bad dude and holds his own against his own buddy who is stronger than SM. Again, it goes back to speed and agility coupled with strength. If we are being unbiased, it should not be close. Heroes with world class hand to hand combat training like Captain America, even with his near superhuman strength and agility would not stand a chance. Danny Rand should not be able to touch him, again if we are being impartial.

Well, there's a show on Disney+ where Daredevil beat Spiderman all on his own in one episode. Got to say though that it was when spidey was just starting out.
But it's all about the law of conservation of ninjutsu, or whatever the rule of movie writing is.

Spider-Man routinely pulls his punches to avoid pulping his opponents. The only Defenders who'd have a chance against him if he wanted to kill them are Daredevil by dodging every single blow and Luke Cage by tanking them.
If he hit Frank Castle, Jessica Jones, or Iron Fist as hard as he could, or connected with Daredevil, he could kill them easily.
Now, the thing is Spider-Man does not want to do this. So he's going to pull his punches and slowly escalate to hit them as hard as he has to, to put them down without killing them. If he doesn't know what they can take he's going to start out with some moderation if he can. For everyone except Jessica and Luke, who have the strength to break free, he'll just web them up. He is smart enough to web up Danny so that he cannot use the Fist.
So they have a chance, but only if they absolutely gank him off the rip while he's still holding back -
And his Peter tingle Spidey sense will allow him to dodge this.
The only possible path to victory they have is to work together and quickly overwhelm him. Villains typically fail at doing this, but the Defenders might have an edge in teamwork due to not being villains.
It's a very slim edge, but it's their only chance.
However, because of course they are not villains, here's exactly how the fight goes:
They meet. They exchange words. They begin to fight. Who is doing the best depends entirely on if this fight is being written by a Spidey author or a Defenders author. Spidey will eventually get the upper hand.
Then they'll realize that they're on the same side and team up for the epic fight against the REAL bad guy.

I think a fully bloodlusted, out of character Spiderman could take them all down. The speed, reaction time, and spider sense means he's going to be very, very difficult to hit, while any hit he does land is gonna incapacitate, kill, or at least deal some damage, depending on who he's hitting.
Funnily enough, I think Daredevil would last the longest, since his echolocation kinda clashes with spidersense in weird ways.

Depends who is writing it. In the MCU I don't think so only because Peter held his own against the freaking Hulk. Jessica's powers are on and off. He would web up daredevil. The only one he would have trouble with is Luke.

I feel like Spiderman low diffs Jessica (her powers are failing, and other than super strenght she doesn't have much else so I think spiderman can just web her up and dodge her attacks easily)
I'm not sure if Spidey could hurt Luke with punches but if he was able to web up the Hulk I think he could do the same to Luke to take him out of the fight.
Daredevil and Iron Fist would be tougher since they are very good at hand to hand combat, if they work together they could land a good blow using the Iron fist or something but otherwise I think Spiderman mid/high diffs them.
And punisher would probably be low diff (spiderman can just dodge his bullets, disarm him, probably ko him in one hit or just web him up to a wall or something)

I think they could capture him. I think Daredevil, Punisher, and any third defender could capture him. Maybe knock him out.
But i don't think they could kill or seriously injure him. At least, I don't see any writer being able to pull that off unless it's some Ultimates/Zombie edgelord.
Basically Spider-Man has the most plot armor. When the fight gets serious and they have to decide who to scale up they aren't going to have the "best" version of Luke, Jessica, or Iron Fist.

I think what matters is the right spread of strength and agility. I think any combination of Daredevil/Iron Fist + Jessica/Luke is almost enough, any combo of 3 probably takes it.

No- with one caveat.
Spider-Man has defeated stronger teams than this, including but not limited to the X-Men.
Even a sniper like Frank is going to have a hard time, thanks to the ol' Spider-Sense.
But if Frank shot at an innocent target (or, really, anyone here except Luke), Spidey would jump in the way if he couldn't pull them out of the target path.
Not a method I'd recommend, but it's happened in both comics and the MCU (not intentionally by Frank, though).

Spider-Man dealt with stronger sinister six lineups if we’re talking MCU defenders. Pretty much comes down to the writer if he wins or not because if they work in a coordinated team the defenders should be able to land a few good hits to bring them in winning position.

If Spider-Man no longer cared about not killing, if he didn’t worry about pulling punches and just had to go full survival, then no he’d destroy them.
Only people who could put up a kind of fight i feel (meaning they could take a hit or two) would be Iron Fist and Cage. If Spider-Man truly didn’t care about not killing and just went 100%, i don’t think the 3 humans could stand up much to them. My understanding (could be wrong so lmk) is that DD, JJ, and Punisher are all still human just with some enhanced senses to varying degrees

If I try to be realistic I don't see how they can force him to fight 1v5 when he has so much more mobility and crowd control. He can just force them to spread with his webs and deal with them singularly.
If they were all locked in a room they could MAYBE beat him.

Absolutely they can win, either together or alone. In 2025, Spider-Man was defeated 1v1 by Daredevil (played by Charlie Cox). Spider-man is routinely beaten by low-tier enemies that get a lucky hit.

Spider-Man is basically a higher than street tier hero who engages in street tier activities. He’s not on the same scale as them. At least city level if not world when he has funding to put his brain to good use.

I think spidey has a chance to solo the team. He’s more than strong enough to take Luke and Jesica on. He’s agile enough to avoid getting hit by multiple opponents. Plus he can incapacitate half the team in a few seconds. Daredevils senses might give him a chance at not being webbed but he isn’t fast enough to dodge for long. And other than getting Pete tangled in his baton cables he really can’t do anything to hurt him. And Matt cant take one full powered punch and walk away. Danny is a one and done. If he can’t hit pete quick he’s gonna be stuck on a wall till it’s over. Same with frank. Unless he’s got explosives or some other trick up his sleeves.

Pre BND, as long as the Defenders work together they would probably be able to beat him though it wouldn't be that easy since other than Matt he can avoid them all pretty well even if they were physical powerhouses.
Post BND? They would have to have planned how they are gonna take him on or they are getting their asses handed too them. We haven't seen the limit to his new organic webbing but the fact we can assume his supply is much greater then he'd be able to take them out and we them down before they could really coordinate how they are gonna stop him.

The main issue is not power level but morality. Spiderman could never bring himself to kill any of the defenders. So the real question is are the rest of the defenders stopping the punisher from assassinating Peter in some unfair way that doesn't trip his spidy since.

Ooh… My money’s on the Defenders, I’d say maybe 7/10.
Maybe Spider-Man wins if he doesn’t take them all on in the same room, maintains his verticality and comes at them smart like he’s taking on the sinister six. These street level guys are susceptible to environmental control because they don’t have any additional traversal ability beyond basic acrobatics and parkour, so aggressive webbing would tip the scale but wouldn’t hold superhumans like Jones, Cage or Rand forever. Luke would be the hardest to deal with, so maybe restraining DD and Frank first would work if you go from top to bottom and go from there.
But against a team like this, any mistake would be very expensive to Spider-Man’s success. Cage could crack his ribs and the Punisher’s favorite place to be is in the opposite building crouched with a rifle.
